abstract | (57) [Problem] To provide an enzymatic quantification method capable of simply and accurately measuring homocysteine and cysteine. SOLUTION: The relationship between the cysteine content and the amount of hydrogen sulfide generated is made constant using homocysteine and an enzyme (E2) that reacts with cysteine, and the enzyme (E2) is contained in a sample containing cysteine and homocysteine. Under the conditions, the amount of generated hydrogen sulfide (1) is measured. Next, the amount of hydrogen sulfide derived from cysteine (2) was determined from the cysteine content in the sample separately obtained using the above constant, The homocysteine content in the sample is determined by subtracting it from the amount of hydrogen sulfide (1). The cysteine content can be determined by measuring the amount of hydrogen sulfide generated by allowing an enzyme (E1) that specifically reacts with cysteine to generate hydrogen sulfide to act on the sample. |
