titleOfInvention Manganese peroxidase with excellent resistance to hydrogen peroxide
abstract PROBLEM TO BE SOLVED: To provide a manganese peroxidase excellent in hydrogen peroxide resistance, which can be used in the technical field of decomposing or degrading lignin such as pulp bleaching in the pulp and paper industry and the like. SOLUTION: Ganoderma strain YK-505 (FERM) P-16991) and have the following properties: manganese peroxidases G-1 and G-2. (A) Action and substrate specificity: oxidizes 2,6-dimethoxyphenol and guaiacol in the presence of hydrogen peroxide and divalent manganese ions. Further, the unbleached pulp is whitened in the presence of hydrogen peroxide and divalent manganese ions. (B) Stability against hydrogen peroxide: shows a residual activity of 30% or more (35% for G-2) in the presence of 10 mM or less of hydrogen peroxide. (C) Optimum pH and pH stability: The optimum pH is 4.0 and stable in the range of pH 4.0 to 7.0 (G-2 is pH 4.0 to 7.5). (D) Optimum temperature and temperature stability: The optimum temperature is 55 ° C and is stable up to 60 ° C. (E) Molecular weight: 44,000 (G-2 is 52,000) (By SDS polyacrylamide electrophoresis)
