abstract | Preparation of 4-(6-methoxy-2-naphthyl)-2-butanone (I) (Nabumetone) comprises condensing 6-methoxy-2-naphthaldehyde with furfuryl acetoacetate, followed by the hydrogenolysis and decarboxylation of the resulting 3-furfuryloxycarbonyl-4-(6-methoxy-2-naphthyl)-3-buten-2-one (II). Preferably the condensation is carried out in the presence of basic catalysts, preferably piperidinium acetate. The hydrogenolysis is carried out in the presence of palladium-on-charcoal, and directly on the condensation reaction mixture, without recovering compound (II). |
