abstract | Compsn for determining the Mg concn in an aq liq contains glycerol kinase (GK); alpha-glycerol phosphate oxidase (GP0); a cpd (I) with peroxidase-equivalent activity; ATP; glycerol; a peroxidase-oxidisable cosubstrate (II); an indicator-cosubstrate coupler (III); a buffer to maintain pH 5.5-9.2, and water. The buffer is Tris-HCl, and the compsn is formulated as 2 reagent solns; one contains glycerol and (III), the other contains all other components. (III) are benzidine, p-phenylenediamine or p-aminophenol cpds; 4-aminoantipyrine (AAP); 3-methyl-2-benzothiazolinone or their mixts, and (III) is at least one NH4 or alkali metal salt of 2-hydroxy-3,5-dichlorobenzene sulphonic acid (IIIa). GK is an Mg-dependent enzyme from Streptomyces canus or Bacillus stearothermophilus, and GP0 is derived from Aerococcus viridans. |
