abstract A method for preparing monomeric calicheamicin derivative/carrier conjugates with higher drug loading/yield and decreased aggregation of the formula Pr(-X-S-S-W)m(I) wherein: Pr is a proteinaceous carrier radical, X is a linker that comprises a product of any reactive group that can react with a proteinaceous carrier, W is the calicheamicin radical formed by removal of the naturally occuring methyl trisulfide group; and m is a number from 0.5 to 15, said method comprising the steps of: (1) incubating a calicheamicin derivative and a proteinaceous carrier in a non- nucleophilic, protein-compatible, buffered solution having a pH in the range from about 4.0 to 8.5 which solution further comprises (a) a cosolvent selected from the group consisting of propylene glycol, ethanol, DMSO, and combinations thereof; and (b) an additive comprising at least one C6-C18 carboxylic acid, wherein the incubation is conducted at a temperature ranging from about 25!C to about 37!C for a period of time ranging from about 15 minutes to about 24 hours to produce a calicheamicin derivative/carrier conjugate; and (2) purifying the calicheamicin derivative/ carrier conjugate produced in step (1) to produce a monomeric calicheamicin derivative/carrier conjugate of formula I. 1311 א' בתמוז התשס" ד - June 20, 2004
