abstract | 1490466 Detection of fibrinogen &c. using dyed Staph. aurens WARNER-LAMBERT CO 17 Feb 1976 [7 March 1975] 06135/76 Heading GIB A compositions for determining fibrinogen, fibrin split products and fibrinogen split products in blood comprises an imiazole buffer suspension of killed Staphylococcus aurens cells dyed with a triphenyl tetrazolium chloride dyestuff or with fast Black Salt K. (a diazo-salt of 2, 5-dimethoxy-4- [(4-P-nitrophenyl)-azo] -benzenamine). The dyestuff may be 2, 3, 5-triphenyl tetrazolium chloride, 2- (P-iodophenyl)-3-(P-nitrophenyl)-5-phenyltetrazolium chloride or 3, 31-dimethoxy-4,41- biphenylene)-bis-(2, 5-diphenyl-2H-tetrazolium chloride). A saline solution of the dyestuff may be added to the fermentation broth in the presence of a polyalkylene glycol molecular weight from 1000 to 5000, then the killed dyed cells are washed free of unused dyestuff. When fast Black Salt K is used it is added to a suspension of the killed cells. The killed dyed cells suspended in imidazole buffer may be lyophilized. In use in the determination of fibrinogen etc.; aliquots of the dyed cells are added to serial dilutions of the serum under test in wells of a microtitre plate and the wells inspected for clumping of the cells. |
