abstract | 1331959 Preparing 6-aminopenicillanic acid BIOCHEMIE GmbH 25 Nov 1970 [28 Nov 1969] 55981/70 Heading C2A [Also in Division C3] 6-Aminopenicillanic acid is prepared by treating a penicillin of formula or an alkali metal salt thereof, wherein R is benzyl, phenoxymethyl, phenylmercaptomethyl, p-cresoxymethyl or n-butoxymethyl, with a penicillin amidase prepared by aerobically cultivating Bovista plumbea (NRRL 3501) or a penicillin amidase-producing mutant or variant thereof in a nutrient aqueous medium. The amidase may be in the form of the whole culture, the filtrate or isolated mycelium, an acetone powder of the mycelium or a partially purified enzyme extract. Also the mycelium, preferably as an acetone-dried powder, or a purified enzyme preparation may be mixed with an inert carrier, e.g. chalk, silica aerogel, diatomaceous earth or sea sand, and formed into a granulate. Particles of dried mycelium or the said granulate may be coated with a permeable film of polymeric material, e.g. polyacrylic resin. These various solid particulate forms of the amdiase may be used to treat the aforesaid penicillins by column methods, or in batch procedures in a fermentor tank.
