abstract | Compounds are of the class of 1-substituted-4-aryl-2(1H)-quinazolinones, e.g., 1-ethyl-4-phenyl-2(1H)-quinazolinone, which are useful as anti-inflammatory agents, antipyretics and analgesics. The compounds are prepared by any one of several processes including, inter alia, the cyclization of a 2-alkylaminobenzophenone with urethane, the cyclization of a 2-alkylaminobenzophenonimine with phosgene, the reaction of a 1-alkali metal salt of a 4-aryl-2(1H)-quinazolinone with an alkyl halide or from a 1-unsubstituted-4-arylquinazoline which is reacted with a methyl halide to form the corresponding 1-methyl-quinazolinium halide which is either then directly oxidized or first reduced and then oxidized to obtain the 1-methyl-4-aryl-2(1H)-quinazolinone. Novel intermediates disclosed include the 1-methyl-4-aryl-quinazoliniums and compounds which are 1-methyl-4-aryl-3,4-dihydroquinazolines.n[US3723432A] |
