[Also in Division A5] Novel dibenzodiazepines of Formula (I) (in which R 1 is H or C 1-4 alkyl, each of R 2 and R 3 is H or halogen, and R 4 is a monocyclic, 5 to 7 membered, heterocyclic ring linked to the carbon atom of the acetyl group via a nitrogen atom, the ring, if desired, containing a cyclic oxygen atom or a further cyclic nitrogen atom and/or being substituted by one or two C 1-4 alkyl, C 1-4 hydroxyalkyl or C 1-4 alkoxy groups or by a benzyl or methylbenzyl group) and acid addition salts thereof, are prepared by reacting a compound of Formula (II) (in which Hal is a halogen atom) with a compound of formula R 4 H, and, if desired, converting a resulting compound I into an acid addition salt thereof. Compounds I in which R 4 represents a 1-piperazinyl group unsubstituted in the 4-position may also be prepared by hydrogenolysis of a corresponding compound I (prepared as described above) in which R 4 represents a 4-benzyl-1-piperazinyl group. Starting materials of Formula (II) are prepared by reacting a corresponding 5-unsubstituted dibenzodiazepine with chloroacetyl chloride. 2 - Chloro - 5,10 - dihydro - 10 - methyl - 11H- dibenzo - [b, e][1,4] - diazepin - 11 - one is prepared by reacting the potassium derivative of 2 - chloro - 5,10 - dihydro - 11H - dibenzo- [b, e][1,4] - diazepin - 11 - one with methyl iodide.n[GB1236112A] |
