abstract | High purity D-(-)penicillamine is prepd. by reacting naturally occurring penicillin salts e.g. benzyl- or phenoxy-methyl-penicillin, or penicillin acids e.g. phenoxy-methyl-penicillin with hydrazinium-hydrochloride, acetic-acid and hydrochloric-acid at 20-120 deg.C for 1-30 hrs. with constant stirring in an inert solvent with b.pt. below 120 deg.C contg. no or less than 20wt.% water and removing the water from the reaction mixt. during or at the end of the reaction by azeotropic distillation. - The reaction prod. is sepd. from the reaction mixt. by filtration, washed with aliphatic alcohol, mixed with ethanol and treated with hydrochloric-acid and filtered. The filtrate is purified with charcoal and treated in a nitrogen atmosphere in the presence of an equiv. amt. acetic-acid with a base, pref. diethyl-amine. The resulting crystalline prod. is filtered, washed with aliphatic alcohol and dried. |
