abstract | Beta-lactam antibiotics (I) having an alpha-formamido substit. on the C atom adjacent to the CO gp. of the ring are new. They are esp. cpds. of formula (II) or their salts. In (II), R1 is H, an acyl gp. or an amino-protecting gp.; R2 is H or a readily removable carboxyl-protecting gp.; Y is -S-CMe2-, -S-CH2- or -Y1-CH2-CZ: Y1 is O, S or CH2; Z is H, halogen or an organic gp. such as 1-4C alkoxy, CH2Q or CH:CHQ; and Q is H, halogen, OH, SH, CN, COOH, carboxylic ester, 1-4C alkoxy, acyloxy, aryl, heterocyclyl bonded via a C atom, hetercyclylthio or a N-contg. heterocyclyl bonded via a N atom. Cpds. (I) are broad-spectrum antibacterials, esp. when R1 is acyl in cpds. (II), and they have esp. good activity against Gram-negative bacteria and are useful as therapeutic agents in man and animals. Other cpds., esp. (II; in which R1 is H or a protecting gp.) are intermediates in the prepn. of the N-acyl derivs. |
