titleOfInvention | Indole derivatives for use in the treatment of central nervous system diseases, pharmaceutical compositions containing them and their use |
abstract | The present invention relates to dopamine and serotonin receptor ligands of formula (I). In formula (I), (a) Y1 and Y2 are one of the nitrogen atoms attached to Y4 and the other of Y1 and Y2 is CO, CS, SO or SO2 and Y4 is CH2; (b) one of Y1 and Y2 is a nitrogen atom attached to the substituent Y4 and the other of Y1 and Y2 is CH2 and Y4 is CO, CS, SO or SO2; or (c) one of Y1 and Y2 is a nitrogen atom attached to the substituent Y4 and the other of Y1 and Y2 is CH2 and Y4 is CH2; Y3 is Z-CH2, CH2-Z or CH2CH2, and Z is oxygen or sulfur, with the proviso that when Y1 is nitrogen, Y3 is other than Z-CH2; W is a chemical bond or an oxygen or sulfur atom or a CO, CS, SO or SO2 group; n is an integer from 0 to S, m is an integer from 0 to 5, and m + n is an integer from 1 to 10, provided that when W is oxygen or sulfur, then n is equal to or greater than 2 and m is equal to or greater than 1; if W is CO, CS, SO or SO2, the value of akkorn is equal to or greater than 1 and m is equal to or greater than 1; X is CH or carbon or nitrogen, with the proviso that when X is carbon, it denotes a dotted line chemical bond, and when X is nitrogen or CH, the dotted line does not represent a chemical bond; R 1 -R 9 are independently selected from hydrogen, halogen or cyano, nitro, amino, hydroxy, alkylamino, dialkylamino, alkyl, alkenyl, alkynyl, alkoxy, alkylthio, hydroxy or thiol; cycloalkyl, cycloalkylalkyl, acyl, thioacyl, aryl, trifluoromethyl, trifluoromethylsulfonyl or alkylsulfonyl; and R10 is hydrogen, alkyl, alkenyl, alkynyl, hydroxy or thiol substituted alkyl, cycloalkyl, cycloalkylalkyl, aryl, arylalkyl, acyl, thioacyl, alkylsulfonyl, trifluoromethylsulfonyl or arylsulfonyl. The compounds of the invention may be used to treat various psychiatric and neurological disorders such as schizophrenia, other psychoses, anxiety disorders, depression, migraine, cognitive disorders, ADHD and sleep disorders. The invention also encompasses compounds for use in pharmaceutical compositions. HE |
