titleOfInvention | Substituted phenylpiperazine derivatives, pharmaceutical compositions containing them and their use |
abstract | The present invention relates to novel substituted phenylpiperazine derivatives capable of binding effectively to 5-HT1A receptors, to pharmaceutical compositions containing such compounds, and to pharmaceutical compositions for the treatment of certain psychiatric and neurological disorders of the compounds of the invention. Many of the compounds of the present invention act as effective serotonin reuptake inhibitors and / or D3 / D4 ligands, particularly useful for the treatment of depression and psychosis. In the present invention, the phenylpiperazine derivatives of the present invention are represented by the following formula: wherein Z is oxygen or sulfur or imino or -NR 3 'and R 3' is hydrogen or alkyl; R7 and R8 are independently hydrogen or halogen, or alkyl, cycloalkyl, cyano, trifluoromethyl, or alkoxy, or form a fused 5 or 6 membered aryl or heteroaryl group for the benzene ring R7 and R8; Y is nitrogen or carbon or methine; the dotted line refers to a chemical bond present in a particular case; R6 and R6 'represent a hydrogen atom or an alkyl group; X is O or S; n is 2, 3, 4 or 5; m is 2 or 3; R 1, R 2, R 3, R 4 and R 5 are independently hydrogen or halogen or alkyl, alkenyl, alkynyl, cycloalkyl, aryl, hydroxyl, hydroxyalkyl, alkoxy, cycloalkoxy, alkylsulfanyl, acyl or -NR 9 R 10 in general, R9 and R10 are independently hydrogen or alkyl, alkenyl, alkynyl, cycloalkyl or aryl, or R9 and R10 taken together with the nitrogen to which they are attached, 1-morpholinyl, 1-piperidinyl, 1 -homopiperidinyl, 1-piperazinyl, 1-homopiperazinyl, 1-imidazolyl, 1-pyrrolyl or pyrazolyl, and any of the mentioned groups may be optionally substituted with alkynyl; or R 1, R 2, R 3, R 4 and R 5 are two-membered condensed with the phenyl ring to form a cyclic group of formula (XVII), (XVIII), (XIX) or (XX), wherein W is oxygen or sulfur, and R 'is R'; and R <is hydrogen or alkyl.
