The present invention relates to a process for preparing rapamycin 31-silyl ether by reacting (a) rapamycin with a silylating agent to give rapamycin 31,42-bis-silyl ether and (b) obtaining 31,42-bis-silyl ether. hydrolyzed with cold, dilute acid to yield rapamycin31-silyl ether.
