Mutants of Mycobacterium species and their use for screening antibiotic vectors |
abstract |
The present invention relates to novel Mycobacterium species mutants and their use in screening methods for identifying potential antibiotic vectors. The novel mutants of the invention are characterized in that the genes encoding components of the siderophore-mediated iron uptake system are inactivated. Gene inactivation shall be carried out by one of the following methods: (a) mutagenesis by chemical and / or physical mutagens; (b) <gene exchange <, that is, replacement of the entire gene or part of the gene with a marker gene; (c) deletion or insertion inactivation; or (d) a combination of the above methods. The mutants thus obtained can be used in methods for screening for siderophores, which can be transported into the Mycobacterium cell independently of the exocelin / mycobactin transport system and thus are considered potential antibiotic vectors.
