abstract | The present invention provides a human / mouse chimeric antibody comprising a murine monoclonal antibody variable (V) region against human tissue factor (TF) and a human antibody constant (C) region. The invention further provides a humanized antibody comprising the complementarity determining regions (CDRs) of the light chain (L chain) variable region and the heavy chain (H chain) variable region (V region) of the same antibody monoclonal antibody when grafted into a human antibody. The invention further relates to the L-chain and the H-chain of said antibody; a V region fragment forming part of the L chain and the H chain, and a method for producing an anti-humanized antibody to human tissue factor. The invention further relates to DNA molecules encoding the antibodies of the invention, recombinant vectors carrying the DNA of the invention, and host cells transformed with the vectors of the invention. The invention further provides a method of producing a chimeric antibody and a humanized antibody against human tissue factor. The invention also relates to a pharmaceutical composition and a therapeutic agent for the combined treatment of disseminated intravascular coagulation (DIC) symptoms comprising an anti-humanized antibody of human tissue factor as an active ingredient.
