abstract | The present invention relates to a process for the preparation of modified epothilones at position 16.17. The process consists of either protecting the 3.7-protected or unprotected epothilones A and H at position 3,77 by either a) hydrogenation of the 16,17 double bond, or b) halogenation of the 16,17 double bond, or c) They are epoxidized at the 16,17-double bond and optionally reduced to the 16-alcohol. The invention further relates to a process for the preparation of epothilone N-oxides, wherein the corresponding epothilones A or B are converted to N-oxides and optionally O-alkylated.
