abstract The present invention relates to a process for the preparation of doxifluridine of formula (I) in which 2 ', 3'-O-isopropylidene-5-fluorouridine of formula (II) is reacted with a sulfonic acid of formula (III) R-SO3H (III) - wherein R is reacted with an alkyl derivative of alkyl, trifluoromethyl, phenyl or mono-, di- or trisubstituted phenyl; (b) reacting the 5 '- (sulfonyloxy) derivative of the general formula (IV) thus obtained with an alkali metal or alkaline earth metal iodide; (c) hydrolyzing the resulting 5'-deoxy-5'-iodo-2 ', 3'-O-isopropylidene-5-fluorouridine of formula (V) in an acidic medium; and (d) reducing the 5'-deoxy-5'-iodo-5-fluorouridine of formula (VI) with hydrogen or a hydrogen donor.
