titleOfInvention | ENANTIOSELECTIVE PREPARATION OF QUINOLINE DERIVATIVES |
abstract | Process for the preparation of 8-substituted oxy - ((R) -2-halo-1-hydroxy-ethyl) - (1H) -quinolin-2-one or acceptable solvates thereof, comprising the reaction of 5- (α-haloacetyl) ) -8-substituted oxy- (1H) -quinolin-2-one with a reducing agent in the presence of a chiral agent and a base to form 8- (substituted oxy) -5- (R) -2-halo-1-hydroxy -ethyl) - (1H) -quinolin-2-one, wherein said chiral agent has the formula I or II wherein M is Ru, Rh, Ir, Fe, Co or Ni; L is a C6-C24-aryl or C6-C24-aryl-C1-C10-aliphatic residue, which in both cases is optionally attached to a polymer; X is hydrogen or halo; R1 is a C1-C10-aliphatic, C3-C10-cycloaliphatic, C3-C10-cycloaliphatic-C1-C10-aliphatic, C6-C24-aryl-C1-C10-aliphatic residue or a 4- to 12-membered heterocyclic group which is in in each case optionally linked to a polymer; and R2 and R3 are phenyl; or R2 and R3 together with the carbon atom to which they are attached form a cyclohexane or cyclopentane ring.
