titleOfInvention PROCEDURE FOR THE PRODUCTION OF TNFR-IG FUSION PROTEIN
abstract A method for producing a tumor necrosis factor and immunoglobulin (TNFR-Ig) receptor fusion protein in bulk cell culture, comprising the steps of: obtaining a cell culture comprising: mammalian cells comprising a gene encoding the TNFR-Ig gene which is expressed under cell culture conditions; glutamine-containing imedium having the characteristics of a medium selected from the group consisting of: (i) a cumulative amount of amino acids per unit volume greater than about 70 mM, (ii) a molar ratio of cumulative glutamine to cumulative asparagine of less than about 2, (iii ) molar ratio of cumulative glutamine to cumulative total amino acids less than about 0.2, (iv) molar ratio of cumulative inorganic ions to cumulative total amino acids between about 0.4 and 1, (v) combined cumulative amount of glutamine and asparagine per unit volume about 16 mM, and combinations thereof; maintaining said culture in the initial growth phase, under a first set of culture conditions, during a first period of time sufficient to allow said cells to reproduce to a vital cell density within the range of about 20% -80% of the maximum possible vital cell density, when said culture is maintained under the first set of cultural conditions; modifying at least one culture condition so that a second set of culture conditions is then applied; maintaining said culture during the second period of time under this second set of conditions, and in the second period of time TNFR-Ig accumulates in the cell culture.
