abstract |
The present invention relates to complex metal transition compounds, with an emphasis on 99mTc and Re, which exhibit high blood-brain barrier permeability so that they can be used as diagnostic / therapeutic agents for CNS diseases. The compounds are of the general formula wherein the M (CO) 3+ tricarbonyl metal nucleus is complexed with a cyclopentadienyl anion attached to heterocyclic compounds of the benzothiazole, benzimidazole and benzoxazole families. The crossing of the blood-brain barrier for three compounds with M = 99mTc is between 7.04 and 3.99% ID / gr in 2 min. In addition, the compounds bind in vitro to β-amyloid peptide fibrils and to amyloid plaques characterizing Alzheimer's disease, inhibit β-amyloid peptide toxicity in cell cultures, and produce ROS associated with Alzheimer's disease. They therefore have proven potential for application in the preparation of diagnostic / therapeutic preparations for Alzheimer's disease. In addition, the high crossing of the blood-brain barrier makes them candidates for investigation for the preparation of drugs for CNS diseases. |
