titleOfInvention Improvements in or relating to antibiotics and their preparation
abstract A new antibiotic designated 9865 RP is produced by cultivating aerobically Streptomyces 8899 (NRRL 3046) or Streptomyces 31723 (NRRL 3045) or a 9865 RP productive mutant thereof in a nutrient medium containing assimilable sources of carbon, nitrogen, and mineral salts until substantial antibiotic activity is produced by the micro-organism in the medium, and recovering the antibiotic from the medium. 9865 RP is an amorphous orange-red powder soluble in chloroform and dichloroethane, moderately soluble in water acidified to pH 3-4, very slightly soluble in alcohols and practically insoluble in water, benzene and diethyl ether. It contains carbon, hydrogen, oxygen and nitrogen in the proportions C = 61.0%, H = 6.2%, O = 28.15%, N = 2.65%. Three constituents, of which 13057 RP and 13213 RP are described, the third being designated 13330 RP, may be isolated from 9865 RP by chromatography or counter-current distribution. 13057 RP is a base which forms acid addition salts, the free base being an amorphous red powder soluble in alcohols and chloroform, very slightly soluble in water and ketones and practically insoluble in benzene and diethyl ether. It contains carbon, hydrogen, oxygen and nitrogen in the proportions C = 59.8%, H = 5.85%, N = 2.3%, O = 29.45% and melts at 155-170 DEG C. (with decomposition). 13213 RP is an amorphous orange-red powder soluble in chloroform and dimethylformamide, moderately soluble in water acidified to pH 3-4, very slightly soluble in alcohols and practically insoluble in water, benzene and diethyl ether. It contains carbon, hydrogen and nitrogen in the proportions C = 58.6%, H = 6.2%, N = 2.2% and also contains oxygen. The Specification contains particulars of the ultra-violet and infra-red spectra of 9865 RP, 13057 RP, 13057 RP hydrochloride, 13213 RP and the aglycone of 9865 RP. A pharmaceutical composition comprises at least one of the antibiotics 9865 RP, 13057 RP and 13213 RP or non-toxic acid addition salt thereof, in association with a compatible pharmacologically acceptable carrier.
