abstract The invention comprises the alkaloid harmidine and the corresponding phenol harmidol, and the preparation of harmidine from Peganum harmala by (i) percolating powdered seeds thereof with methanol or ethanol, and removing the alcohol, (ii) partitioning the residue between water and petroleum ether, (iii) treating the aqueous phase with ammonia and ammonium sulphate, (iv) precipitating crude hydroiodides with potassium iodide solution, (v) recrystallising, (vi) liberating the free bases with alkali and dissolving them in acetic acid, (vii) adding ammonia and heating so as to hydrolyse the acetate of the weaker base harmine, and filtering off the harmine formed, and (viii) adding ammonia to hydrolyse harmidine acetate, and recrystallising the product. Harmidine may be converted (a) into harmidol by treatment of the hydroiodide with ammonia, (b) into dihydroharmidine by treatment with zinc and hydrochloric acid, and (c) into dibromoharmidine by treatment with bromine; the monobromo-derivative is obtained by treating the dibromo-derivative with ammonium hydroxide. Salts of harmidine and harmidol are also disclosed.
