titleOfInvention Enteric-coated tablets of dextran sulphate ester and method of preparation thereof
abstract The invention comprises a tablet of a water-soluble salt of dextran sulphate coated uniformly with an enteric coating material which is insoluble in a simulated gastric fluid having a pH below 3.0 but dissolves or disintegrates in a simulated intestinal fluid having a pH ranging between 5.0 and 8.0. The coating may be 0.05 to 1.0 mm. thick and is preferably followed by a further coating of acetoglyceride (an acetylated aliphatic glyceride wherein the carbon atoms of the fatty acid are 12 to 20) and a wax such as beeswax, carnauba wax, vegetable wax or synthetic wax. Typical coating materials are sodium, potassium and ammonium salts of alginic acid; sodium potassium and ammonium salts of cellulose acetate phthalate; sodium, potassium and ammonium salts of cellulose acetate maleate; sodium, potassium and ammonium salts of polyvinyl alcohol phthalate; and sodium, potassium and ammonium salts of polyvinyl alcohol maleate; cellulose acetate phthalate, cellulose acetate maleate, polyvinyl alcohol phthalate, and polyvinyl alcohol maleate. The dextran sulphate e.g. sodium or potassium salt, is mixed with a diluent such as starch and compressed or moulded to form a tablet. The tablet is coated with an adhesive e.g. aqueous gelatin and the enteric coating then applied. The operation may be repeated to give the desired coating. To assist dispersion of the dextran sulphate in the intestine and to promote absorption from the intestine the dextran sulphate salt may be mixed with isopropyl myristate and the stearic acid ester of a polyoxyethylene prior to granulation and compression into tablets. The tablets of the invention are effective in the treatment of hyperlipemia.
