titleOfInvention | Improvements in or relating to lysergic acid derivatives |
abstract | The invention comprises lysergic acid derivatives of general formula: <FORM:0950528/C2/1> in which R1 is a C1-4 alkyl radical, each of R2 and R3 is a hydrogen atom, a C1-4 alkyl or hydroxyalkyl radical or a monocyclic aryl radical, and \sG signifies the radical -CH=C< or <FORM:0950528/C2/2> ; and their acid addition salts; and a process for preparing these compounds in which the corresponding lysergic acid azide is heated to form the isocyanate, which is reacted with an amino compound <FORM:0950528/C2/3> , and, if desired, salification is effected with an organic or inorganic acid. The azides are prepared from the corresponding lysergic and dihydrolysergic acid hydrazides using hydrochloric acid and sodium nitrite solution, and the hydrazides in turn are prepared by heating 1-substituted derivatives of natural ergot alkaloids, or their dihydro derivatives, with anhydrous hydrazine. In the examples the amines used in the preparation of the carbamides are anilene, diethylamine and (+)-butaolamine -(21). Pharmaceutical compositions, having sedative, serotonin inhibiting and/or blood pressure lowering action, comprise the compounds of the invention together with inert carriers. Specification 948,203 is referred to. |
