titleOfInvention Process for the production of substituted tetrazoles
abstract 1-Substituted tetrazoles substituted in the 5-position by an organic radical containing an a -hydroxy group are obtained by reacting an isonitrile with an aldehyde or ketone and hydrazoic acid or an azide; the isonitrile provides the 1-substituent, and the aldehyde or ketone R COR1 becomes the group RR1C(OH)-attached to the 5-position of the tetrazole ring. Specified isonitriles are fulminic acid and its salts, and isopropyl, butyl, t.-butyl, cyclohexyl benzyl, pyridyl and 2, 6-dimethylphenyl isocyanide; specified carbonyl compounds are formaldehyde, acetaldehyde, propionaldehyde, butyraldehyde, isobutyraldehyde, benzaldehyde, ferrocene-aldehyde, pyridine-aldehyde, benzthiazole-aldehyde, succin-dialdehyde, 1-azulen-11-propen -1- al, propinal and the b -amino acroleins obtained therefrom by addition of amines, dimethylaminocinnamaldehyde, glycerinaldehyde and other aldoses, b -methylmercaptopropionaldehyde, trifluorobutyraldehyde, b -dimethylaminopiral-aldehyde, cyclohexanone, and dimethyl-, methylethyl-, diethyl-, methylpropyl-, ethylpropyl-, dipropyl-, dibutyl-, benzylmethyl- and dibenzylketones. Examples prepare the tetrazoles substituted in the 1- and 5-positions respectively by: (1) 2,6-dimethylphenyl, hydroxymethyl; (2) cyclohexyl, 1-hydroxy -2- methylpropyl; (3) cyclohexyl, 1 - hydroxy - 2,2,2 - trichloroethyl; (4) 2,6-dimethylphenyl, 1-hydroxy-2,2,2-trichloroethyl; (5) p-diethylaminophenyl, 1-hydroxy-2,2,2-trichloroethyl; (6) cyclohexyl, 1-hydroxy -1- methylethyl; (7) cyclohexyl, phenyl hydroxymethyl; (8) cyclohexyl, 1-phenyl -1-hydroxyethyl; (9) benzyl, 1-hydroxy-2,2-dimethyl -3- dimethylaminopropyl; and (10) 2,6-dimethylphenyl, 1-hydroxy -2, 2-dimethyl -3-dimethylaminopropyl.
