titleOfInvention Amino-compounds of the oestrane series and a process for their manufacture
abstract The invention comprises 3-amino-17b -hydroxy-D 1,3,5(10)-oestratriene and the corresponding derivatives in which the 17b -hydroxyl group is functionally converted, for example, an aceoxy group, and a process for the preparation of 3-amino-compounds of the oestrane series wherein a p-quinol of the said series of the general formula <FORM:0938432/IV(a)/1> wherein R1 represents hydrogen or an acyl radical and in which the non-aromatic portion of the steroid system can include any substituent which takes no part in the reaction, is reacted with a hydrazine derivative of the formula RNHNH2 in which R represents an organic group such as a phenyl or 2,4-dinitrophenyl group or a group of the formula NH2CO-, and the resulting azodyestuff is reductively split up, for example, by means of dithionite or hydrogen in the presence of a hydrogenation catalyst. The reactant for the above process may be a p-quinol derived from oestradiol or a 17-ester thereof or from oestrone or a 17-protected keto derivative thereof. The process may be illustrated by the following reaction scheme <FORM:0938432/IV(a)/2>ALSO:Azodyestuffs are prepared by reacting a p-quinol of the oestrane series having the general formula <FORM:0938432/IV(a)/1> wherein R1 represents hydrogen or an acyl group and the non-aromatic portion of the steroid system can include non-reactive substituents such as a 17b -hydroxy or -esterified hydroxy group or a 17-keto or -protected keto group, with a hydrazine derivative of the formula RNHNH2 wherein R is an organic group such as a 2,4-dinitrophenyl or phenyl group or a group of the formula NH2CO-. The process is illustrated by the following reaction scheme. <FORM:0938432/IV(a)/2>
