abstract The invention comprises compounds having the formulae: p <FORM:0923945/IV(a)/1> <FORM:0923945/IV(a)/2> wherein X is hydrogen, fluorine or chlorine and R1 is b -hydroxymethylene or carbonyl and the 1,2-carbon linkage is a single or double bond linkage, and a process for the preparation thereof which comprises reacting a compound of the formula: <FORM:0923945/IV(a)/3> wherein X has the value given above, Y is b -hydroxymethylene or carbonyl and R is iodine or an alkyl- or aryl-sulphonyloxy radical, or the corresponding D 1,4-unsaturated compound with a metal fluoride. Said compounds may be obtained for example by esterifying the 21-hydroxy group of 6a ,16a -dimethyl-11b , 17a ,21-trihydroxy-4-pregnene-3,20-dione or the corresponding D 1,4 compound with an alkyl or aryl sulphonyl chloride to produce the corresponding 21-sulphonate ester which is treated with an alkali metal iodide to produce a 21-iodo compound, which gives the 21-fluoro compound on treatment with silver fluoride. Illustrative products of the invention are 6a , 16a - dimethyl - 11b ,17a - dihydroxy-21-fluoro-1,4-pregnadiene-3,20-dione, 6a ,16a -dimethyl-9a ,21 - difluoro-11b ,17a -dihydroxy-1,4-pregnadiene-3,20-dione and the corresponding 4-pregnene compounds.ALSO:Pharmaceutical compositions comprise a compound having the formula <FORM:0923945/VI/1> wherein X is hydrogen, chlorine or fluorine atom, R1 is b -hydroxymethylene or carbonyl and the 1,2-carbon linkage is a single or double bond linkage, and a pharmaceutical diluent. They are administered orally as pills, tablets, syrups and elixirs, in liquid forms for injection, and topically as ointments, creams, sprays and lotions, with or without coacting antibiotics, e.g. the penicillins, neomycin, tetracyclin, chloramphenicol and novobiocin.
