titleOfInvention 6ª--fluoro-21-methyl-í~-pregnadien-17ª--ol-3,20-dione intermediate therefor and preparation thereof
abstract The invention comprises 6a - and 6b -fluoro21-methyl-D 4,9(11)-pregnadien-17a -ol-3, 20-diones, the preparation of the 6b -compound by reacting a 6b -fluoro-21-methyl-D 4-pregnene-11,17a -diol-3,20-dione 11-alkyl or aryl-sulphonate with an alkaline reagent such as an alkali metal salt of a fatty acid containing up to 4 carbon atoms or an alkali metal halide, and the isomerization of the 6b -compound to the 6a -compound by treating it with a prototropic agent at a temperature between -5 DEG and 10 DEG C. The reaction with the alkaline reagent is preferably conducted in an anhydrous solvent, the sulphonate being of the 11a - or 11b -series. Suitable prototropic agents are water, alcohols and organic and mineral acids. Examples are given. 6b -Fluoro - 21 - methyl-D 4-pregnene-11,17a diol-3,20-dione 11-alkyl or aryl-sulphonates are prepared by reacting 21-methyl-D 5-pregnene - 3b ,17a - diol - 20 - one with N - bromo acetamide and hydrogen fluoride to give 5a -bromo - 6b - fluoro-21-methyl-pregnane-3b ,17a diol-20-one, oxidizing this to the corresponding 3-ketone, heating this with an alkaline reagent to give 6b -fluoro-21-methyl-D 4-pregnen17a -ol-3,20,dione, microbiologically hydroxylating this to give a 6b -fluoro-21-methyl-D 4pregnene-11,17a -diol-3,20-dione and esterifying this with a sulphonyl halide. Examples of these processes are also provided.
