titleOfInvention Lysergic acid-alkaloids
abstract The invention comprises two isomeric derivatives of D-lysergic acid and D-isolysergic acid having a carbinolamidic structure and empirical formula C18H21O2N3, which form maleates C22H25O6N3, yield acetaldehyde and D-lysergic acid amide or D-isolysergic acid amide on hydrolysis and are probably of the formula <FORM:0883330/IV (b)/1> also a process for their preparation by filtering a culture broth produced by the fermentation in submerged culture of virulented strains of Claviceps paspali Stevens and Hall, and extracting the filtrate with an organic solvent at a pH between 7 and 9 and at a temperature between 0 DEG C. and 10 DEG C. The extract is then concentrated and purified by successive transfers from the organic solvent (e.g. chloroform) and an acidic aqueous phase (e.g. tartaric acid solution) between 0 DEG C. and 10 DEG C., adjusting the pH of the aqueous phase to pH 7-9 before each extraction with the organic solvent. Treatment of either of the new compounds with acid (or with alkali) yields an equilibrium mixture of the two. Examples detail (a) the preparation of D-lysergic acid amide, D-isolysergic acid amide and small amounts of the new compounds (using normal temperature extraction) and (b) isolation of the new compounds by extraction at 0.5 DEG C. and equilibration of the individual isomers with aqueous acetic acid.
