titleOfInvention New hydrazinium compounds, their production and pharmaceutical compositions containing them
abstract The invention comprises compounds of the formula <FORM:0882154/IV (b)/1> (wherein R is phenyl, thienyl or furyl which may be substituted by halogen or alkoxy of at most 7 carbon atoms; R1 is phenyl, pyridyl or pyrimidyl; R2 and R3 are alkyl radicals each containing at most 7 carbon atoms, A is an alkylene radical of 2 to 6 carbon atoms; and X is an anion bearing the charge n) and their acid-addition and quaternary ammonium salts. They may be prepared by reacting chloramine or hydroxylamine-O-sulphonic acid with the appropriate tertiary amine, or by reacting an unsymmetrical dialkyl hydrazine with a compound of the formula RCH2N(R1)AX. Anions may be exchanged by metathesis. Examples describe the preparation of 1 : 1-dimethyl-1-(N-2-thenyl-N-2-pyridyl-2-aminoethyl)-hydrazinium chloride, picrate, picrate hydrogen picrate, benzylpenicillinate, 5 : 5-dimethyl - 3 - pyrazolyl - 4 - (N - methyl) - aminomethanesulphonate; 1 : 1 - dimethyl - 1 - [N - (2 - pyrimidyl) - N - (p - methoxybenzyl) - aminoethyl] hydrazinium acid sulphate, iodide- and o-acetoxybenzoate; 1 - 1-dimethyl-1-(N-phenyl-N-benzylaminoethyl) hydrazinium sulphate and gluconate; 1 : 1-dimethyl-1-[(N-benzyl-N-2-pyridyl)-2-aminoethyl] hydrazinium chloride, chloride hydrochloride, picrate hydrogen picrate, 2-carbamoyl-phenolate, nitrate and acetate; and 1 : 1-dimethyl-1-(N-(2-pyridyl)-N-(5-chloro-2-thenyl)-aminoethyl) hydrazinium chloride. Other salt forming acids are also referred to, and an extensive list of suitable anions X is given. Specificatio 858,488 and U.S.A. Specifications 2,678,258 and 2,710,248 are referred to.ALSO:Pharmaceutical compositions comprise a compound of the formula <FORM:0882154/VI/1> (wherein R is phenyl, thienyl or furyl which may be substituted by halogen or alkoxy of at most 7 carbon atoms; R1 is phenyl, pyridyl or pyrimidyl; R2 and R3 are alkyl radicals each containing at most 7 carbon atoms; A is an alkylene radical of 2 to 6 carbon atoms, and X is an anion bearing the charge n) or an acidaddition salt or quaternary ammonium derivative thereof, and a solid or liquid pharmaceutical carrier. Specific examples of X are those listed in Specification 880,974 (except hexafluorophosphate), and gluconate and phenyldimethylpyrazolonylmethylaminomethanesulphonate. Specification 858,488 and U.S.A. Specifications 2,678,258 and 2,710,248 also are referred to.
