titleOfInvention Steroid compounds and the preparation thereof
abstract The invention comprises a compound having the formula (including the D 1, D 6 and D 16 analogues thereof): <FORM:0870387/IV (b)/1> wherein R1 is hydrogen or methyl, R2 is hydrogen, fluorine or methyl, R3 is hydrogen or hydroxyl, R4 is hydrogen, fluorine, methyl, methylene, hydroxyl or acylated hydroxyl, the acyl group containing only carbon, hydrogen and oxygen and being derived from a mono or dicarboxylic acid containing up to 10 carbon atoms, at least three of R1, R2, R3 and R4 always being hydrogen, X is hydrogen, a -halogen, a -methoxy, or a -ethoxy and Y is keto or b -hydroxyl, and a process for preparing compound having the Formula I (including the D 1, D 6 and D 16 analogues thereof) which comprises reacting with diazomethane in the presence of solvent the corresponding 21-aldehyde in anhydrous or monohydrate form. The reaction may be effected in an inert solvent at 0-25 DEG C. Illustrative products of the invention are 17b -(a ,b -oxido-propanoyl) -D 4-androstene11b -17a -diol-3-one, the corresponding D 14 and D 146- compounds and the 9-fluoro and 9-bromo derivatives thereof. Specifications 870,388, 870,389 and 870,390 are referred to.ALSO:Pharmaceutical compositions comprise, in admixture with a pharmaceutically acceptable carrier, a compound of the formula <FORM:0870387/VI/1> (or a D 1-, D 6- or D 1,6-analogue thereof), wherein R1 is hydrogen or methyl, R2 is hydrogen, fluorine or methyl, R3 is hydrogen or hydroxyl, R4 is hydrogen, fluorine, methyl, methylene, hydroxyl or acylated hydroxyl, the acyl group containing only carbon, hydrogen and oxygen and being derived from a mono or dicarboxylic acid containing up to 10 carbon atoms, at least three of R1, R2, R3 and R4 always being hydrogen, X is hydrogen, a -halogen, a -methoxy or a -ethoxy, and Y is keto or b -hydroxyl. The compositions may be in the form of tablets, aqueous suspensions, elixirs, ointments and salves. Specifications 870,388, 870,389 and 870,390 are referred to.
