titleOfInvention Derivatives of novobiocin
abstract The invention comprises methyl 3-carbamyl-4-methylnovobioside and 3-carbamyl-4-methylnovobiose having respectively the formulae : <FORM:0853592/IV(b)/1> and the preparation thereof by reacting novobiocin with a non-oxidizing mineral acid in methanolic solution to produce the former compound with the optional further step of reacting the product with a dilute aqueous nonoxidizing mineral acid solution to produce the latter compound. A suitable non-oxidizing mineral acid is hydrochloric acid. The invention also includes (a) a process for the preparation of 3-carbamyl-4-methylnovobiose diethyl mercaptal of the formula : <FORM:0853592/IV(b)/2> by reacting methyl 3-carbamyl-4-methylnovobioside with ethyl mercaptam in the presence of hydrogen chloride with optional further items of reacting the product with Raney nickel to produce 3-carbamyl-4-methyl-1-deoxynovobiose of the formula: <FORM:0853592/IV(b)/3> hydrolysing this compound with sodium hydroxide to form 4-methyl-1-deoxynovobiose of the formula: <FORM:0853592/IV(b)/4> and treating this compound with carbamyl chloride to form 3-carbamyl-4-methyl-1-deoxynovobiose and 2-carbamyl-4-methyl-1-deoxynovobiose ; and (b) a process for the preparation of methyl 4-methylnovobioside of the formula: <FORM:0853592/IV(b)/5> by heating methyl 3-carbamyl-4-methylnovobioside with sodium hydroxide with the optional further step of reacting the product with carbamyl chloride to form methyl 3-carbamyl-4-methyl-novobioside of the formula given above and methyl 2-carbamyl-4-methylnovobioside. Detailed examples are given. Specifications 821,819 and 821,820 are referred to.
