titleOfInvention | Improvements in or relating to steroids and the manufacture thereof |
abstract | The invention comprises a compound having the general formula <FORM:0822118/IV (a)/1> wherein R13 is a hydroxy, acyloxy or keto group, wherein the acyl radical is of a hydrocarbon carboxylic acid containing from 1-8 carbon atoms inclusive, and R111 is an alkyl or aryl radical, a compound having the general formula <FORM:0822118/IV (a)/2> wherein R1 is hydrogen or the acyl radical of a hydrocarbon carboxylic acid containing from 1-8 carbon atoms inclusive, and the corresponding 3,20-bis-[alkylene ketals] thereof, wherein the alkylene radical contains not more than 8 carbon atoms inclusive and the attaching oxygen atoms are separated by a chain of at least 2 and not more than 3 carbon atoms inclusive; a compound having the general formula <FORM:0822118/IV (a)/3> wherein R1 and the ketal groups are as above defined; and a process for the preparation thereof which comprises treating a compound having the general formula <FORM:0822118/IV (a)/4> with an organic peracid to obtain the corresponding 5a , 6a -oxido-11a -acyloxy-allopregnane - 3,20 - dione - 3,20 - bis - [alkylene ketal], treating the latter compound with an alkyl or aryl metal compound to obtain the corresponding 5a ,11a - dihydroxy - 6b - alkyl or aryl - allopregnane - 3,20 - dione 3,20 - bis - [alkylene ketal] treating the latter compound with dilute acid to obtain a 5a , 11a -dihydroxy-6b -alkyl or arylallopregnane-3,20-dione, dehydrating with an alkali metal base to obtain 6-alkyl or 6-aryl-11a -hydroxyprogesterone and then if desired oxidizing to produce the corresponding 11-keto derivative. The 6a -compound is obtained by treating the compound thus obtained with an alkali metal base. The examples are particularly directed to the preparation of the 6-methyl compounds.ALSO:6 - Methyl - 11a - hydroxyprogesterone, 6 - methyl - 11 - keto - progesterone and other 6-alkyl or aryl steroids are employed for the treatment of arthritis in the form of tablets and injectables, for the treatment of topical inflammatory conditions of the skin in the form of ointments or for the eyes, ears and nose in the form of drops, nasal sprays or ointments. They have sedative and hypnotic activity and potentiate the sleep-producing activity of hexobarbital. |
