abstract | A therapeutic composition comprises a water-insoluble penicillin salt in finely-divided form, polyvinyl-pyrrolidone in amount from about 0.05 to 2.0 per cent, lecithin in amount from about 0.2 to about 5.0 per cent, a parenterally acceptable water-soluble buffer in amount from about 0.5 to 10 per cent, an alkali metal salt of carboxymethylcellulose in amount from about 0.1 to 3 per cent, all by weight of the penicillin salt, and a preservative. Specified penicillin salts include procaine penicillin, dibenzylethylenediamine penicillin; N-benzyl-N1 - (3 - ethoxy - 4 - hydroxy) - benzylethylenediamine penicillin; 2-chloroprocaine penicillin and N,N1 - di - (4 - methoxybenzyl) - ethylenediamine penicillin. Polyvinylpyrrolidone of average molecular weight 40,000 is preferred. Suitable buffers include sodium citrate, sodium acetate, sodium lactate and alkali metal salts of inorganic acids such as mono- and di-basic sodium phosphates. Specified preservatives include the methyl, propyl and butyl esters of p-hydroxybenzoic acid, sodium bisulphite, and quaternary ammonium compounds such as benzalkonium chloride. A stable polyhydroxy alcohol such as sorbitol or mannitol may also be included. A minor proportion of a solubility depressant (e.g. procaine hydrochloride when procaine penicillin is employed, or dibenzylethylenediamine acetate when dibenzylethylenediamine penicillin is employed) may also be included in the compositions. The dried preparation is suitable for extemporaneous compounding with water to form aqueous suspensions for injection purposes. An example is included. |
