titleOfInvention Substituted androstanes and androstenes and the preparation thereof
abstract The invention comprises 3b -acyloxy-17b -hydroxy-17a -ethinyl androstanes wherein the acyl group is an alkanoyl group having from 3 to 10 carbon atoms such as a butyryl, valeryl, caproyl or heptanoyl radical or any of the radicals listed below, or a carboxyalkanoyl group having from 4 to 6 carbon atoms, 3b -acyloxy - 18b - hydroxy - 17a - ethinyl - 5 - androstenes wherein the acyl group is a propionyl, isobutyryl, 2-ethyl-n-butyryl, isovaleryl, trimethylacetyl, isocaproyl, octanoyl, nonanoyl or decanoyl group, or a carboxyalkanoyl group having from 4 to 6 carbon atoms such as a b -carboxypropionyl, g - carboxybutyryl or d - carboxyvaleryl radical, and 3b -cycloalkylacyloxy-17b -hydroxy - 17a - ethinyl - androstanes and 5-androstenes wherein the cycloalkyl group is a 5-6-membered ring which may be substituted by an alkyl group having from 1 to 4 carbon atoms and the acyl group is an alkanoyl group having from 1 to 8 carbon atoms, and the preparation of 3b -acyloxy-(or 3b -cycloalkylacyloxy) - 17b - hydroxy - 17b - ethynyl - androstanes or 5-androstenes by reacting the required 3b ,17-diol compounds with alkanoic or cycloalkanoic acylating agents, the alkanoic group being derived from an alkanoic acid having at least 3 carbon atoms and the cycloalkyl ring being a 5- or 6-membered ring which may be substituted by an alkyl group having from 1 to 4 carbon atoms. The alkanoic acylating agent may be an alkanoic anhydride or acid halide having from 3 to 10 carbon atoms or the anhydride of an alkanedioic acid having from 4 to 6 carbon atoms. The cycloalkylalkanoic acylating agent may be a cycloalkylalkanoic anhydride or acid halide to form compounds in which the radical in the 3-position may be a cyclopentyl- or cyclohexyl-substituted formyloxy or acetoxy radical, a - or b -cyclopentylpropionoxy, a - or b -cyclohexylpropionoxy, b - or g -cyclopentylbutyroxy, g - cyclohexylbutyroxy, cyclohexylisobutyroxy, d - (2 - methylcyclohexyl) valeryloxy, o - cyclohexylvaleryloxy, g - and o - cyclohexylcaproyloxy, o - (3 - ethylcyclopentyl) caproyloxy, o - cyclohexylheptanoyloxy, o - cyclopentyloctanoyloxy or b - (4 - methylcyclohexyl) propionoxy radical. A large number of examples are given in which the radicals in the 3-position are included in the above list. Reference has been directed by the Comptroller to Specification 765,871.
