abstract | The invention comprises 2,4-diaminoquinazolines of the formula <FORM:0806772/IV (b)/1> (wherein R1, R2, R3 and R4 are hydrogen atoms or lower alkyl groups, at least one being a lower alkyl group and the total number of carbon atoms in all the alkyl groups present being one to five), and the preparation thereof by (a) reacting the corresponding 2 : 4-dihalogenoquinazolines with ammonia or (b) treating the corresponding 2 : 4-dihydroxyquinazoline with a halide or oxyhalide of phosphorus or a mixture of two such compounds to produce a 2,4-dihalogenoquinazoline and reacting the latter with ammonia. In examples, 6-methyl-2,4-dihydroxyquinazoline is reacted with a mixture of phosphorus pentachloride and phosphoryl chloride to form the corresponding 2,4-dichloro compound which in turn is treated with alcoholic ammonia to form 6-methyl-2,4-diaminoquinazoline; similarly is prepared 2,4-diamino-6,7-dimethylquinazoline. In further examples, 2,4-diamino-8-methyl-, and 2,4-diamino-6,8-dimethylquinazolines are prepared in like manner, phosphoryl chloride in dimethyl aniline being used in the chlorination step, and 2,4-diamino-6-ethyl-and 2,4-diamino-5-isopropyl-8-methyl quinazolines are similarly prepared using phosphoryl chloride alone. |
