abstract The invention comprises 5-amino-2:4-dimethyl-pyrimidine and its preparation by condensing acetamidine or a salt thereof with an a -alkoxymethylidene-acetoacetic acid ester for example ethyl a -ethoxymethylideneacetoacetate and treating the resulting 2:4-dimethylpyrimidine-5-carboxylic acid ester by known methods for the conversion of a carboxylic ester into an amine to give the desired compound. The condensation of the acetamidine or salt thereof with the a -alkoxymethylidene-acetoacetic ester is suitably carried out in aqueous or alcoholic solution in the presence of an alkaline agent such as potassium carbonate or an alkali-metal alkoxide at about 20 DEG C. The conversion of the 2:4-dimethylpyrimidine carboxylic or ester to the amine may be effected by the Hofmann degradation, by treating with ammonia to form 2:4-dimethylpyrimidine-(5)-carboxamide, heating with bromine and sodium methoxide to form methyl 2:4-dimethylpyr-imidyl-(5)-carbamate and hydrolysing, for example by aqueous sodium hydroxide or barium hydroxide, to give the amine. Alternatively the Curtius degradation may be used. Examples describe the preparation of the ethyl ester of 2:4 - dimethylpyrimidine - (5) - carboxylic acid and its conversion to 5-amine-2:4-dimethylpyrimidine.
