abstract | A composition in tablet form for use in testing urine or other body fluids for albumin, consists of a mixture containing a protein precipitant, a dye indicator changing colour below pH 6, and an effervescent couple consisting of a bicarbonate and an acid. Protein precipitants used are (1) strong, solid, water-soluble acids, e.g. sulphosalicylic acid or picric acid; (2) sodium tetra- or hexa-metaphosphate; or (3) water-soluble salts, e.g. lead nitrate, sulphate, barium chloride, aluminium sulphate or aluminium chloride. The acid used in the effervescent couple may be an acid of group (1) above, or other strong organic acid, e.g. tartaric, maleic, itaconic, sulphamic, or citric acid. Where the protein precipitant used is an acid or alkali meta phosphate, the albumin is precipitated below its iso-electric point and the dye-indicator used should change colour at pH 2 to 2.5, e.g. Brilliant Green, Crystal Violet, or Cresol Red. When metal salts are used, the alubin is precipitated above the isoelectric point, and the dye-indicator used should change colour near pH 6, e.g. Bromcresol Green, or Diazoresorcinol. The components are mixed together with a little boric acid and calcium sulphate and formed into tablets. In use, one or two drops of urine are placed on the tablet. If albumin is present, the effervescence throws up the precipitated albumin and a colour change results. In a negative specimen forming soon subsides with no colour change. |
