titleOfInvention Morphine derivatives
abstract The invention comprises compounds of the general formula <FORM:0786157/IV (b)/1> in which R is lower (1-5C) alkyl or aryl and R1 is acyl (including carbalkoxy and carbaralkoxy), and the preparation thereof by the action of esterifying agents on the corresponding 6-lower-alkyl- or 6-aryl-dihydromorphine, which in turn may be prepared by reacting dihydromorphinone with an ethereal solution of an alkyl lithium or an aryl lithium. The esterification is advantageously carried out at or slightly above room temperature in the presence of an acid-binding agent (e.g. pyridine) and a solvent (e.g. pyridine, excess of the esterifying agent or a hydrocarbon, halogenated hydrocarbon, ether, ketone or tertiary amine). The esters may be purified by extraction into a solvent immiscible with the reaction mixture, and subsequently crystallized by trituration with ether. Examples describe the interaction of: 6-methyldihydromorphine (prepared from dihydromorphinone and methyl lithium) with (1) methyl chlorocarbonate, (2) ethyl chlorocarbonate, (5) benzyl chlorocarbonate and (6) acetic anhydride; and of ethyl chlorocarbonate with (3) 6-ethyldihydromorphine (from dihydromorphinone and ethyl lithium), (4) 6-phenyldihydromorphine (from dihydromorphinone and phenyl lithium; the hydrochloride is described), and (7) 6-butyl-dihydromorphine (from dihydromorphinone and butyl lithium).
