titleOfInvention Improvements in or relating to the production the antibiotic griseofulvin
abstract Griseofulvin, an antifungal antibiotic useful in agriculture, is produced by the submerged aerobic fermentation of a griscofulvia-producing organism in a culture medium containing an assimilable source of nitrogen at a level of 0.04 to 0.30, and preferably 0.075 to 0.25, per cent available nitrogen, a source of carbon and energy and nutrient salts. The nitrogen source may be sodium nitrate but preferred are complex organic nitrogen containing materials such as corn steep liquor, cottonseed meal, oatmeal, soya bean meal and milk products such as whey powder, buttermilk and liquid whey. The carbon source is preferably a carbohydrate especially lactose, glucos, sucrose or starch, used in quantities of at least 3.5 and preferably 5.0 per cent. The nutrient salts include: (a) phosphates, e.g. potassium mono- and dihydrogen phosphate in quantities of 0.4 to 0.8 per cent; (b) calcium carbonate in quantities of 0.4 to 1.2 per cent to buffer the fermentation broth; (c) chloride ions in the form of soluble chlorides, e.g. potassium chloride in quantities of at least 0.1 per cent to prevent the formation of dechloro griseofulvia; and (d) magnesium ions. The medium is initially adjusted to pH 4.5 to 5.5 and is preferably inoculated with a vegetative inoculum in proportions of 1 to 10 per cent of the medium, the inoculum being prepared with a medium containing less than 0.25 per cent available nitrogen. The fermentation temperature is 25 DEG C. and aeration is good. Fermentation is stopped before the mycelium breaks down and the griseofulvin is recovered from the mycelium by extraction with an organic solvent such as ethyl acetate, amyl acetate, butyl acetate or n-butanol. The extract is concentrated in vacuo before 50 DEG C., the solid removed, washed with solvent and purified by dissolving in a solvent such as acetone and precipitating the griscofulvin by the addition of water. The medium used in the example contains corn steep liquor solids 0.2 per cent, lactose 7 per cent, limestone 0.8 per cent, potassium dihydrogen phosphate 0.4 per cent and potassium chloride 0.1 per cent. The organism is Penicillium patulum Bainier Thom (4640, 455) C.M.I. 39,809 and this strain or mutants thereof is preferred, although other griseofulvin-producing organisms are specified.
