titleOfInvention Isoalloxazines
abstract The invention comprises isoalloxazines of the general formula <FORM:0764339/IV(b)/1> wherein n is 2-6, R1 is either an acyloxy radical in which the acyl group is derived from an organic acid containing not more than 18 carbon atoms, or a sulphate radical, or a phosphate radical, R3 and R4 are hydrogen or halogen atoms, or alkyl or alkoxy radicals containing up to 5 carbon atoms or amino groups, or methylene groups linked together to form a carbocyclic ring containing 5 or 6 carbon atoms, R2 and R5 are hydrogen atoms, or alkyl or alkoxy radicals containing up to 5 carbon atoms, or amino groups, and wherein R2, R3, R4 and R5 when taken together include not more than one amino group; and their preparation by reacting the corresponding alcohol with an acid chloride or acid anhydride of sulphuric acid or phosphoric acid or with an organic acid containing up to 18 carbon atoms or a chloride or anhydride thereof. When the latter are used a base such as sodium carbonate or pyridine is preferably added, but when the free acid is used the reaction is preferably carried out by refluxing in the presence of an acid catalyst such as a mineral acid or toluene sulphonic acid. Examples describe the preparation of 6,7-dimethyl-9-isoalloxazine ethyl acetate, dihydrogen phosphate, hydrogen phosphate and phosphate, hydrogen sulphate and sulphate, propionate, hydrogen succinate and mixed succinate esters such as methyl succinate, succinate, benzoate, laurate, palmitate, b -cyclopentane propionate, salicylate, hydrogen phthallate and mixed esters such as methyl phthallate, and phthallate; similar esters of the following 9-[b -hydroxyethyl]-isoalloxazines: 7-methyl-, 6 - ethyl - 7 - methyl -, 6,7 - dichloro -, 6-methyl-7-amino- and 6,7-diethyl-; similar esters of 6,7-dimethyl-9-[g -hydroxypropyl]-isoalloxazine, including the myristate; 5-methoxy-6-amino-9-isoalloxazineethyl propionate; 5 - amino - 6 - oxy-9-isoalloxazine-ethyl acetate; 6-methoxy-8-amino-9-isoalloxazine-ethyl acetate and 6-amino - 8 - methoxy - 9 - isoalloxazine-ethyl acetate. A further list of possible products is given together with a list of various acids from which the products of the invention are derived, including hydroxy carboxylic acids, amino carboxylic acids, sulphonic acids and phosphonic acids. Specification 760,068 is referred to.
