titleOfInvention | Improved process for the recovery of serum albumin |
abstract | Serum albumin is recovered from an aqueous solution of mammalian serum containing other protein constituents of mammalian blood by selectively coagulating the other protein constituents by heating the solution to a temperature between about 45 DEG and 75 DEG C., the solution being at a pH on the alkaline side of the isoelectric point of the serum albumin but not above 5.7 and containing caprylate ions at a concentration of between approximately 0.0075 and 0.02 M. The aqueous solution should preferably contain not more than 12 per cent solids. The caprylate ions are generally introduced by adding a water-soluble salt of caprylic acid to the solution. The pH may be adjusted to required value, preferably about 4.9 to 5.5, by means of acids or acid salts such as hydrochloric, sulphuric and phosphoric acids or their acid salts and acetic or oxalic acid. The denatured globulin and other non-albumin proteins are removed after the heating and purified albumin recovered from the residual solution by evaporation to dryness, if desired after dialysis, or precipitation by means of a water-miscible organic solvent. Examples describe the recovery of serum albumin from bovine plasma, hog plasma, lyophilized sheep plasma and bovine whole blood. Blood serum may also be used. The aqueous solutions treated may contain small amounts of lower alcohol as when whole blood is treated with ethanol in 4 per cent concentration at pH about 7.3 to remove fibrinogen. |
