abstract | The invention comprises an 11b ,17a - dihydroxypregnan-20-one having the formula <FORM:0738611/IV (b)/1> wherein Ac is hydrogen or an acyl radical of an unsubstituted monocarboxylic acid containing from one to eight carbon atoms inclusive, and the preparation thereof by mixing together a reducing agent and a 17a -hydroxypregnane-11,20-dione 20-ketal, having a substituent at the 3-position of the formula AcO, and wherein the ketal group contains the polymethylenedioxy radical of an alkane-1,2-diol or alkane-1,3-diol containing from two to eight carbon atoms, inclusive, to produce a 3-hydroxy- or 3 - acyloxy - 11b ,17a - dihydroxypregnane - 20 - one-20-ketal, and hydrolysing the product by means of an aqueous acidic hydrolysing agent. The reduction may be effected by lithium aluminium hydride, sodium borohydride, lithium borohydride or by hydrogenation in the presence of a catalyst, e.g. platinum, palladium on charcoal or Raney nickel. The hydrolysis may be effected with a mineral acid. Examples disclose the preparation of 3a - and 3b -11b ,17a -trihydroxypregnan-20-one, 3a - and 3b - acetoxy - 11b ,17a -dihydroxypregnan - 20 - one and 3a - benzoyloxy - 11b ,17a - dihydroxypregnan-20-one. Specification 737,701 is referred to. |
