titleOfInvention Improvements in or relating to the production of therapeutically effective sulphonamide compounds
abstract 550,040. Sulphonamido compounds. LILLY & CO., E., KHARASCH, M. S., and REINMUTH, O. Nov. 6, 1940, No. 16181. [Class 2 (iii)] Therapeutically useful compounds are prepared by reacting formaldehyde and an unsubstituted mercapto-carboxylic acid or mercapto-sulphonic acid with 4 - p - aminobenzenesulphonamide or a derivative thereof containing in the amide group an hydroxyl group, an hydroxylated lower alkyl group having more than one carbon atom, an # - pyridyl group, a 2 - thiazolyl group or a substituted 2 - thiazolyl group, preferably in the presence of a non-oxidising mineral acid. The products may be converted into their salts with alkali and alkaline earth metals, ammonia, alkylamines, alkanolamines and polymethylenediamines. Examples are given of the treatment of (1) p - aminobenzenesulphonamide, p - aminobenzenesulphonylethanolamide and # - p - aminobenzenesulphonamidopyridine with formaldehyde and thioglycollic acid, and the conversion of the products into their sodium, ammonium, ethanolamine, diethanolamine, propanolamine, isopropanolamine and ethylenediamine salts ; (2) 2 - p - aminobenzenesulphonamido - 4 - methylthiazole, 2 - p - aminobenzenesulphonamidothiazole, 2 - p - aminobenzenesulphonamido - 4 - phenylthiazole and 2- p - aminobenzenesulphonylhydroxamide with formaldehyde and thioglycollic acid, and the conversion of the products into their sodium salts ; (3) p - aminobenzenesulphonamide with formaldehyde and p - mercaptobenzenesulphonic acid, oc - p - aminobenzenesulphonamidopyridine with formaldehyde and # - mercaptopropionic acid, 2 - p - aminobenzenesulphonamido - 4 - methylthiazole with formaldehyde and o - mercaptobenzoic acid, and the conversion of the products into their sodium salts.
