abstract 501,799. Materials for diagnosing tuberculosis. EVERY, C. E. (Lakeland Foundation) June 18, 1937, Nos. 17056/37 and 5230 /39 (the second Provisional Specification being divided out of the first Provisional Specification). [Class 81 (i)] An agent specific to tuberculosis, which on injection produces a reaction indicative of the disease when present, is prepared by extracting a protein which is characteristic of tuberculosis from the solid unaffected tissue or from the blood fibrin of an animal suffering from the disease and making an approximately neutral or mildly acid solution thereof. The extracting agent may be a dilute acid or alkaline solution, e.g. of hydrochloric acid, or sodium hydroxide. The solid tissue used may be fibrous connective tissue, muscular tissue or bone marrow. In obtaining the agent from fibrin, blood is coagulated, the clot washed free from haemoglobin and serum, hardened with acetone and dried.; the dried fibrin is then pulverized and extracted with decinormal sodium hydroxide, centrifuged, and the liquid rendered neutral or slightly acid by the addition of hydrochloric acid and potassium phosphate, the final pH preferably being 6-9 Tricresol and glycerine is added to the preparation as a preservative. The agent is similarly obtained from muscular tissue by removing blood and fat, drying, pulverizing and extracting.. If tuberculosis is present in a subject, a slight area of inflammation and the formation of pseudopods is observed round the point of injection.
