abstract | Keto - cyclopentanodimethyltetradecahydrophenanthrols are prepared from pregnanolones by a process which comprises converting the latter into secondary-tertiary dialcohols, splitting out water from the tertiary hydroxyl group and the adjoining hydrogen atom, followed by a removal of the side chain at the double bond by the action of oxidizing agents characterized in that the removal of water is accomplished without simultaneous acylation of the secondary alcohol group, and preferably by a dry heating of a Grignard reaction product with excess of a Grignard reagent, for example in a high vacuum under reflux. The splitting up of the double bond is preferably carried out by means of ozone, but other oxidizing agents such as permanganate, and chromium trioxide may be used. When ozone is used, the ozonide is preferably split by a reduction method, using a platinum catalyst, and shaking an ethyl acetate solution in a hydrogen atmosphere. In an example, allo-pregnanol-(3)-one-(20) is treated with magnesium methyl iodide. Water is split off from the tertiary alcohol so produced by heating at 110 DEG C. for one hour under reflux in a high vacuum. The dehydrated product is treated in chloroform solution with ozone, and the ozonide decomposed by heating at 50 DEG C. with glacial acetic acid to give the required hydroxy - ketone - (3 : 17). If, however, the ozonide is split by shaking with a platinum catalyst in ethyl acetate solution in an atmosphere of hydrogen, there is obtained the corresponding di-secondary di-alcohol. Specification 449,326 is referred to. A sample has been furnished under Sect. 2 (5) of trans-androsterone made as follows. Allo-pregnanol - (3) - one - (20) is treated with magnesium methyl iodide, the product dehydrated by heating in a vacuum under reflux, and then treated with ozone. The ozonide on heating with glacial acetic acid gave the required product. |
