abstract | Quaternary derivatives of 3-aminoacridines are manufactured by treating 3-aminoacridine or such substitution products thereof as contain, in the benzene ring not substituted by the amino group, halogen, alkyl or alkoxy groups, all of which compounds are substituted in the amino group by a substituent capable of being split off, such as an acyl group, with an alkylating agent, and subsequently splitting off the substituent from the amino group. The products are intended for use against blood parasites. The 3-aminoacridines employed are obtainable by the process of Specification 367,024. In examples: (1) 3-aminoacridine is treated with acetic anhydride, the acetyl derivative methylated by means of p-toluene-sulphonic acid methyl ester and the product saponified with hydrobromic or hydrochloric acid, yielding 3-amino-10-methylacridinium bromide or chloride; (2) 3-amino-7-methylacridine, 3-amino-6-methylacridine and 3-amino-7-chloroacridine are similarly converted into 3-amino-7 : 10-dimethylacridinium chloride, 3-amino-6 : 10-dimethylacridinium chloride and 3-amino-7-chloro-10-methylacridinium chloride respectively; (3) 3-amino-7-ethoxy-10-methylacridinium chloride and 3-amino-6 : 7-dimethoxy-10-methylacridinium chloride are similarly prepared; (4) 3-amino-7-ethyl-10-methylacridinium chloride is similarly produced. |
