titleOfInvention Improved manufacture of tuberculin preparations
abstract 285,087. I, G. Farbenindustrie Akt.- Ges. Feb. 11, 1927, [Convention date]. Toxins, vaccines, and immunizing and like serums.-Tuberculin preparations are treated with heavy metal compounds such as dialysed iron oxychloride or lead salts so as to remove albumins and their cleavage products leaving the active constituents in the solution, from which they may be separated by addition, either of acetone, an alcohol, or other solvent in which the active substances are insoluble, or of an alkaloil precipitant which is afterwards eliminated by chemical action. In one example, dry human tuberculin separated from Koch's broth by precipitation with alcohol, or old tuberculin " Koch," is dissolved in water, and dialysed ferric hydroxide added to precipitate albumin. The concentrated filtrate is treated with picric acid and the precipitate decomposed with dilute hydrochloric acid. Acetone is added to the solution obtained so as to precipitate the active substances. In a second example an extract of bovine tubercule bacilli freed from wax and fat is dissolved in water and the solution treated with lead acetate. The filtrate is freed from lead and treated with acetone. Specifications 21927/93 and 1398/10 are referred to. The Specification as open to inspection under Sect. 91 (3) (a) contains also a third example, according to which killed bacilli are extracted with alcohol and the solution evaporated under vacuum. The residual mass is extracted with water and the solution obtained treated as in the second example. This subject-matter does not appear in the Specification as accepted.
