abstract |
A microfluidic device 100 (e.g. an artificial placenta) comprises base 10 containing first 11 and second 12 microfluidic channels separated by porous membrane 20, (preferably porous polycarbonate) which is coated on both sides 21, 22 by a hydrogel (such as collagen) comprises a first monolayer of cells of a first type 30 (e.g. human placental villous endothelial cells, HPVECs) on one side 21 and a second monolayer of cells of second cell type 30 (e.g. syncytial trophoblasts such as choriocarcinoma (BeWo) cells), are placed on the second side 22 of the membrane wherein that second cell type is differentiated into a syncytial structure. A method of fabricating the microfluidic device and a method of testing placental maternal-foetal transfer using the device under physiological flow conditions is also disclosed. |
